The Phantom in the Noise and Validation of 3D EM Reconstructions
نویسنده
چکیده
Single particle reconstructions derived from electron micrographs yield increasing detail of biological molecules and assemblies. However, it is well-known that alignment of pure noise images against a reference can recover that reference on reconstruction [1]. The question is how to distinguish a reconstruction from noise from one from legitimate particle images. Figure 1A shows the estimated resolutions of reconstructions from projection images with defined signal-to-noise ratios (SNRimp). Comparison with reconstructions from aligned gaussian noise images provides a lower bound for the SNR that can be tolerated (open circles in Figure 1A). Increasing the number of images suppresses noise, giving a relationship between the product of SNR with the number of images and the resolution obtained (Figure 1B). Therefore, given the number of images contributing to a reconstruction with a defined SNR, and considering alignment errors (see below), a particular resolution should be expected. Any excess in images used in a reconstruction may signal validity issues.

Validation of 3D EM Reconstructions: The Phantom in the Noise
Validation is a necessity to trust the structures solved by electron microscopy by single particle techniques. The impressive achievements in single particle reconstruction fuel its expansion beyond a small community of image processing experts. This poses the risk of inappropriate data processing with dubious results. Nowhere is it more clearly illustrated than in the recovery of a reference d...
متن کاملFabrication of New 3D Phantom for the measurement of Geometric Distortion in Magnetic Resonance Imaging System
Introduction: Geometric distortion, an important parameter in neurology and oncology. The current study aimed to design and construct a new three-dimensional (3D) phantom using a 3D printer in order to measure geometric distortion and its 3D reproducibility. Material and Methods: In this study, a new phantom ...
متن کاملValidation of Optimum ROI Size for 123I-FP-CIT SPECT Imaging Using a 3D Mathematical Cylinder Phantom
Objective(s): The partial volume effect (PVE) of single-photon emission computed tomography (SPECT) on corpus striatum imaging is caused by the underestimation of specific binding ratio (SBR). A large ROI (region of interest) set using the Southampton method is independent of PVE for SBR. The present study aimed to determine the optimal ROI size with contrast and SBR for striatum images and val...
متن کاملConventional Voxel in Tomographic Reconstruction Based upon Plane-Integral Projections – Use It or Lose It?
Introduction: While the necessity of replacing voxels with blobs in conventional tomographic reconstruction based upon line-integrals is clear, it is not however well-investigated in plane- integral-based reconstruction. The problem is more challenging in convergent-plane projection reconstruction. In this work, we are aiming at utilizing blobs as alternative to voxels. <stron...
متن کاملAdaptive beamforming in row-column addressed arrays for 3D ultrasound imaging
In recent years, to reduce the complexity of implementation, the use of 2D arrays with restricted row-column addressing has been considered for 3D ultrasound imaging. In this paper, two methods of adaptive beamforming based on the minimum variance method are represented in such a way that the computational load is much less than using the full adaptive beamforming method. In both proposed metho...
